M40, a peptide containing the N-terminal galanin sequence (1-13), is an antagonist of the galanin receptor. M40 significantly antagonized galanin-induced feeding behavior at doses approximately equimolar to the active doses of rat galanin. M40 inhibited the binding of (¹²⁵I-Tyr²⁶)-galanin (porcine) in rat hypothalamic membranes with an IC₅₀ value of 15 nM.
Coadministration of M40 with a muscarinic M1 agonist may enhance the efficacy of cholinergic treatments for the cognitive deficits of Alzheimer disease.
