Phenylacetamidomethyl-Resin (PAM-resin)-Linked Boc-Amino Acids
Resins based on polystyrene-divinylbenzene 1%.
The ester bond between the C-terminal amino acid and Merrifield resin turned out to be not sufficiently stable towards the repetitive treatments with TFA during the Boc-SPPS of long peptides. To avoid such losses the phenylacetamido (PAM) handle has been developed to create a more acid-stable peptide-resin linkage. Cleavage is normally effected by treatment with HF.
